Comparedwithconventionaltherapeuticsystems,drugandgenedeliverysystemsoffernumerousadvantages,includingprolongeddurationtime,reducedsideeffects,improveddrugbioactivityandenhancedtherapeuticefficiency.Toenhancethebioavailabilityoftherapeuticsandtodelivertherapeuticagentstoparticulartissuesandcells,theidealdrugandgenedeliverysystemsshouldhavedesiredfunctionalproperties,includingtargetabilityandstimuliresponsibility.Ourstudiesfocusedonthefunctionalpolymersandpeptidesfordrugandgenedelivery.Toendowthedeliverysystemswithactivetargetingproperty,studieswerecarriedoutontheincorporationofvarioustargetingligandsincludingparticularcellpenetratingpeptidesequence,RGDsequence,transferrin,galactose,andfolateetc.todifferentdeliverysystemsthroughcovalentbondsaswellasbiotin-avidininteraction.Togivethedeliverysystemswithstimuliresponsibility,wedesignedvariousdrugdeliverysystemswithstimuliresponsibilitiesincludingpH,temperature,redox,andlightsensitivities.Toachieveenhancedsynergyeffects,double/multiplestimuli-responsivesystemshavealsobeendeveloped.Thephysical/chemicalpropertiesanddrug/genedeliverybehaviorsinvitroorinvivoofprepareddrugandgenedeliverysystemswereinvestigatedindetail.Theresultsshowedthedrugandgenedeliverysystemsexhibitedimproveddrugcontrolledreleasepropertiesandenhancedgenetransfectionefficiencies.Inaddition,thetargeteddeliverytoparticulartissueandtumorcellscouldbeachievedthroughincorporationofparticulartargetingligands.
