Musselssurviveinturbulentseabyrobustunderwateradhesiontoallkindsofwaveandwind-sweptsubstrata,aswellashighlystiffandhighlyextensiblesurfacecoatingsusingaproteinaceousbyssus.Suchremarkablefeaturesmakethebyssusasourceofinspirationforthedevelopmentofnoveladhesives,coatingsandbiomaterialswhichcanbeintroducedinasimilarlyturbulentaqueousenwronmentinthehumanbody.Themusselbyssusresemblesabundleofthreadstippedwithadhesiveplaquesandcontainsroughly25-30differentproteins.Understandingtheadhesionandcohesionmechanismoftheseproteinsisofbothfundamentalandpracticalimportanceinthedevelopmentofbiomedicaladhesivesandcoatings.Ourgrouphasendeavoredtoinvestigatethemolecularandsurfaceinteractionmechanismsofmusseladhesiveproteinsbyapplyingamolecularforceprobetechnique-SurfaceForcesApparatus(SFA).TheSFAstudieswereperformedinvariousconfigurations:(i)aproteinfilmagainstdifferentsubstrata,(ii)aproteinfilmagainstanothersurfaceofthesameordifferentprotein,and(iii)proteinsolutionsconfinedbetweendifferentsubstrata.Itwasfoundthattheadhesionandcohesionofmusselfootproteinsarecriticallyrelatedtothemodifiedprominentaminoacid,L-3,4-dihydroxyphenylalanine(DOPA)orC2_mannosyl_7_hydroxytryptophan(Man70HTrp).Formusseladhesiontovarioussubstrata,DOPAplaystheimportantroleviadiverseinteractionsincludinghydrogenbonding,metalcoordination,cation-7rinteractions.DOPA-richproteinsmfp-3andmfp-5playamajorroleintheadhesionatplaque-substratainterfaces.DOPAalsocontributessignificantlytotheproteincohesionviametalcoordination(e.g.,DOPA_Ca2+,DOPA_Fe3+),cation-7rinteractions(e.g.,DOPA-Lys+).Ourresultsprovidenewinsightsintothedevelopmentofbiomimeticunderwateradhesivesandcoatingsforbiomedicalapplications.
