尹教授長期以來圍繞“熒光拓撲高分子材料的構(gòu)建與功能化”這一高分子材料領(lǐng)域的重要前沿課題,開展了熒光功能高分子的設(shè)計/合成以及生物應(yīng)用的系統(tǒng)研究。熒光小分子化合物一般具有疏水的共軛結(jié)構(gòu),在生物或極性介質(zhì)中容易聚集,導(dǎo)致熒光衰減或猝滅。此外,傳統(tǒng)高分子載體不含有熒光基團,很難在細胞內(nèi)起到標記、示蹤的作用。因此,需要解決的關(guān)鍵科學(xué)問題是既要確保熒光分子具有水溶性、光穩(wěn)定性,又要賦予熒光分子一定的生物相容性、特異功能性。針對以上關(guān)鍵問題,尹梅貞教授將熒光發(fā)色基團與拓撲功能高分子有機結(jié)合起來,提出了將拓撲功能高分子作為外圍殼保護和隔離疏水熒光核的新策略,構(gòu)建了系列核-殼熒光拓撲高分子材料,解決了水溶性與光穩(wěn)定性差的難題。在此基礎(chǔ)上,發(fā)明了新型熒光抗癌藥物分子以及多種特異性熒光分子探針,使其具有“熒光示蹤”和“納米載體”雙功能 ,開創(chuàng)性地建立了將熒光納米載體用于農(nóng)業(yè)害蟲防治新技術(shù),成功地攜帶基 因/藥物/蛋白應(yīng)用于農(nóng)作物害蟲綠色防控等,為避免化學(xué)農(nóng)藥的過度使用打開了綠色生物防治新思路。研究工作帶動了熒光標記以及抗腫瘤研究領(lǐng)域的 發(fā)展,引領(lǐng)化學(xué)與生命醫(yī)學(xué)以及農(nóng)業(yè)交叉學(xué)科研究,具有重要的學(xué)術(shù)價值以及實際應(yīng)用前景。例如,美國國立研究院Nard Kubben研究員,美國約翰霍 普金斯大學(xué)Jan H. Hoh教授,以及德國美因茨大學(xué)Gert O. Pflugfelder教 授,采用尹教授團隊構(gòu)建的熒光分子用于他們的生物學(xué)示蹤等生命科學(xué)研究 。安貞醫(yī)院心血管重大疾病協(xié)同創(chuàng)新中心(2011協(xié)同創(chuàng)新中心)杜杰教授團隊使用尹教授團隊的熒光抗癌藥物分子進行癌癥早期診斷研究。中國農(nóng)業(yè)大學(xué),華南農(nóng)業(yè)大學(xué)等農(nóng)業(yè)高校研究人員使用尹教授提供的熒光高分子材料從事他們各自領(lǐng)域的不同種類昆蟲遺傳學(xué)等研究。
